Overview

Propyphenazone is a small molecule non-steroidal anti-inflammatory drug (NSAID) of the pyrazolone class, structurally related to phenazone. It acts as an analgesic and antipyretic with some anti-inflammatory activity, primarily used for the treatment of pain and fever, such as headache and toothache. Its mechanism of action involves inhibition of cyclooxygenase enzymes (COX), leading to reduced synthesis of prostaglandins responsible for pain, inflammation, and fever[1][3][7][8]. Propyphenazone is often marketed in combination with paracetamol and caffeine for primary headache disorders[4]. It was introduced in 1951 for rheumatic disorders but has since seen restricted use or bans in some countries due to safety concerns such as blood dyscrasias and rare cardiovascular events[4].
