Prosaptide is a synthetic peptide derived from the neurotrophic region of the multifunctional protein prosaposin. It is a recombinant polypeptide with neuroprotective and glioprotective properties, acting as an agonist at G protein-coupled receptors GPR37 and GPR37L1. Prosaptide has been shown to stimulate neuronal survival pathways, prevent hyperalgesia, reduce peripheral neuropathy, and activate anti-apoptotic mechanisms in preclinical models. It was primarily developed for the treatment of neuropathic pain associated with diabetic neuropathy and HIV-associated sensory neuropathies but did not demonstrate efficacy in phase 2 clinical trials. The drug also showed potential protective effects against ischemia-induced hearing loss and was investigated for its ability to inhibit tumor metastasis via stimulation of p53 and thrombospondin-1 (TSP-1) pathways in cancer models[1][4][5][6][8][9].
