VM110, also known as ProSense 750, is a protease-activatable near-infrared fluorescence (NIRF) imaging agent. It is designed to detect and visualize biological processes characterized by high protease activity, such as inflammation and malignancy. The agent consists of a poly-L-lysine polymer backbone grafted with multiple NIR fluorophores that are optically silent in their native state due to self-quenching. Upon intravenous administration, the probe is internalized or localized to sites of disease where specific cysteine proteases—including cathepsin B, L, and S, as well as plasmin—cleave the backbone. This enzymatic cleavage releases the fluorophores from their quenched state, producing a strong fluorescent signal detectable by NIR imaging systems. VM110 has been investigated for the early detection of ovarian and pancreatic cancers via laparoscopic imaging and for the assessment of inflammatory plaque activity in atherosclerosis.
