Drug intelligence / Profile preview
ProSense VM110 (also known as ProSense 750) is a near-infrared fluorescent (NIRF) protease-activatable imaging agent designed to detect and visualize inflammation and tumor-associated proteolytic activity in vivo. Developed by VisEn Medical (later acquired by PerkinElmer, now Revvity), the agent consists of a pegylated poly-L-lysine polymer backbone graft-copolymerized with multiple near-infrared fluorophores that are optically silent (quenched) in their intact state due to close proximity. Upon intravenous administration, ProSense VM110 is selectively cleaved by cysteine proteases—primarily cathepsin B, and to a lesser extent cathepsins L, S, K, V, and D, as well as plasmin—which are highly expressed by inflammatory cells (such as macrophages) and tumor cells. Proteolytic cleavage separates the fluorophores, dequenching them and generating a strong near-infrared fluorescence signal (excitation/emission peak at 750 nm/780 nm) that can be detected using intravascular NIRF-OCT, NIRF-IVUS, or fluorescence molecular tomography (FMT). It has been evaluated in preclinical models of deep vein thrombosis (DVT), atherosclerosis, and stent-induced vascular injury, and has been investigated in a Phase I clinical trial for the detection of microscopic peritoneal tumors in patients with ovarian, fallopian tube, primary peritoneal, or pancreatic cancers.
