A preclinical **tandem CAR-T-cell therapy concept** designed to target both prostate-specific membrane antigen and NKG2D ligands on tumor cells. The construct combines an anti-prostate-specific membrane antigen single-chain variable fragment with the extracellular domain of the natural NKG2D receptor, enabling recognition of prostate-specific membrane antigen-positive tumor cells as well as tumor cells expressing stress-induced NKG2D ligands. This dual-targeting strategy was developed by Celyad Oncology to address antigen heterogeneity and antigen-negative escape in prostate cancer, including metastatic castration-resistant prostate cancer.
