Prostate stem cell antigen-targeted chimeric antigen receptor natural killer cells (PSCA CAR NK cells) are an investigational, allogeneic, "off-the-shelf" cell therapy designed to treat PSCA-positive malignancies, particularly pancreatic cancer. These cells are typically derived from umbilical cord blood and genetically engineered via retroviral transduction to express a chimeric antigen receptor (CAR) specific to the prostate stem cell antigen (PSCA), a glycosylphosphatidylinositol-anchored protein overexpressed in several solid tumors. To overcome the limitations of NK cell persistence and anti-tumor activity in the immunosuppressive tumor microenvironment, these cells are often further engineered to co-express soluble interleukin-15 (sIL-15). This modification enhances their metabolic fitness and survival without the need for exogenous cytokine support. The therapy is designed to be cryopreserved, allowing for immediate administration to patients without the delays associated with autologous manufacturing.
