PROSTVAC-TRICOM is a therapeutic cancer vaccine designed to stimulate the immune system to target and destroy prostate cancer cells. It is a poxvirus-based vector vaccine that uses two viral vectors—recombinant vaccinia (PROSTVAC-V) for priming and recombinant fowlpox (PROSTVAC-F) for boosting—in a heterologous prime/boost regimen. Both vectors are engineered to express human prostate-specific antigen (PSA), which serves as the tumor-associated antigen, along with three T-cell costimulatory molecules (B7.1, ICAM‑1, and LFA‑3), collectively referred to as TRICOM, to enhance immune activation. The primary mechanism of action is induction of robust PSA-specific CD4+ and CD8+ T cell responses against prostate cancer cells expressing PSA, with evidence also supporting antigen spreading—immune responses against other tumor antigens not present in the vaccine[2][4][8][10]. Developed primarily for metastatic castration-resistant prostate cancer (mCRPC), it has also been studied in earlier-stage disease.
