Drug intelligence / Profile preview
PROTAC-FAK (focal adhesion kinase proteolysis-targeting chimera) is a class of targeted protein degraders designed to selectively bind and induce the ubiquitination and subsequent proteasomal degradation of focal adhesion kinase (FAK, also known as PTK2). Unlike traditional small-molecule FAK inhibitors that only block its kinase activity, FAK PROTACs eliminate the entire FAK protein, thereby simultaneously disrupting both its enzymatic kinase-dependent signaling and its kinase-independent scaffolding functions. In preclinical studies of gastrointestinal neuroendocrine tumors (GI-NETs), the specific FAK PROTAC BI-0319 (developed by Boehringer Ingelheim) has been shown to overcome drug resistance, reduce cell viability, and inhibit tumor cell proliferation, migration, and invasion.
