Drug intelligence / Profile preview
PROTAC tau degraders (or tau-directed proteolysis-targeting chimeras) represent an emerging therapeutic class of heterobifunctional small molecules designed to selectively clear pathological tau protein aggregates, a hallmark of Alzheimer's disease and other tauopathies. These chimeric molecules consist of a tau-binding warhead linked to an E3 ubiquitin ligase-recruiting ligand (such as cereblon/CRBN or von Hippel-Lindau/VHL). By bringing the target tau protein into close proximity with the E3 ligase complex, the PROTAC induces polyubiquitination of tau, marking it for degradation via the cellular ubiquitin-proteasome system. This approach aims to reduce toxic tau accumulation, mitigate neurotoxicity, and slow cognitive decline in neurodegenerative diseases.
