Overview

Protectin D1 n-3 (PD1n-3) is a specialized pro-resolving mediator (SPM) belonging to the protectin family of oxygenated polyunsaturated fatty acids. It is an endogenous lipid autacoid enzymatically derived from n-3 docosapentaenoic acid (n-3 DPA). PD1n-3 acts as a potent agonist for several G-protein coupled receptors, including GPR37, ALX/FPR2, and ChemR23/ERV1, which are expressed on macrophages, microglia, and other immune cells. By activating these pathways, PD1n-3 stimulates the resolution of inflammation, promotes the phagocytosis of cellular debris and apoptotic cells (efferocytosis), and counter-regulates the production of pro-inflammatory cytokines without inducing immunosuppression. Preclinical research has demonstrated its therapeutic potential in various models, including epilepsy (reducing seizure frequency), neuropathic pain, chronic itch, and aggressive brain cancers like medulloblastoma and glioblastoma. It is currently an academic research compound with no commercial developer or clinical-stage programs identified.
