Protego Biopharma's oncology program involves the development of small-molecule activators of the unfolded protein response (UPR) pathway. The UPR is a cellular stress response related to the endoplasmic reticulum (ER) that maintains protein homeostasis. In the context of oncology, these small molecules are designed to exploit the high levels of proteostasis stress and protein misfolding inherent in cancer cells. By further activating the UPR, the program aims to overwhelm the cancer cell's adaptive mechanisms, leading to apoptosis or growth inhibition. The program is currently in the lead optimization phase and represents a novel approach to targeting the proteostasis network in malignant tissues, building on research from The Scripps Research Institute.
