Overview

Protein S human is a vitamin K-dependent plasma glycoprotein that functions as a crucial natural anticoagulant in the human body. It acts as a non-enzymatic cofactor for Activated Protein C (APC), enhancing APC's ability to proteolytically inactivate procoagulant factors Va and VIIIa, thereby regulating blood clot formation. Additionally, Protein S serves as a cofactor for Tissue Factor Pathway Inhibitor (TFPI), accelerating the inhibition of Factor Xa. It also directly inhibits Factor IXa and Factor Xa. Beyond its anticoagulant roles, Protein S is involved in inflammation and efferocytosis by acting as a ligand for the TAM (Tyro-3, Axl, and Mer) family of protein kinase receptors. Therapeutic applications of Protein S, often as a component of combination products like prothrombin complex concentrates (PCCs) or as recombinant protein, are being investigated for managing bleeding disorders, reversing anticoagulation, and preventing thrombosis.
