**Protopanaxadiol (PPD)** is an organic triterpenoid compound that serves as the aglycone (non-sugar portion) of protopanaxadiol-type ginsenosides, found in ginseng and notoginseng species[1][2]. PPD is a dammarane-type tetracyclic terpenoid sapogenin and is considered an isoprenoid with a molecular formula of C₃₀H₅₂O₃[2][4]. As a ginsenoside metabolite, PPD is produced by the human intestinal microflora when ginsenosides like Rb1 are ingested[9]. Preclinical studies suggest a broad spectrum of pharmacological activities, including anti-inflammatory, antioxidant, antidiabetic (improving glucose and lipid metabolism, reducing total cholesterol, triglycerides, and low-density lipoprotein cholesterol, and suppressing pro-inflammatory cytokines[3]), anticancer (inhibiting tumor cell proliferation and inducing apoptosis in various human cancer cell lines[5][7]), and potentially antidepressant effects. Clinical development has focused on its antidepressant potential under the brand name Yoxintine, currently in late-stage clinical trials in China[1]. The exact mechanism of action is not fully elucidated, but PPD binds to glucocorticoid and estrogen beta receptors, increases intracellular calcium ion concentration, and may influence anti-inflammatory and metabolic pathways[1]. It is classified as a natural product and is not currently marketed as a conventional pharmaceutical outside investigational settings.
