Overview

Protoporphyrin IX-beta-cyclodextrin conjugate is a covalent polymer-drug conjugate developed for multimodal cancer therapy and imaging. It consists of the photosensitizer protoporphyrin IX (PpIX) covalently linked at both carboxylic termini to the drug carrier beta-cyclodextrin (betaCD). This modification enhances the water solubility of PpIX and promotes its preferential localization within the mitochondria of malignant cells, such as MCF7 breast cancer and DU145 prostate cancer lines. Upon light activation, the PpIX moiety generates reactive oxygen species for photodynamic therapy (PDT), while its inherent fluorescence allows for diagnostic imaging. Furthermore, the cyclodextrin component can encapsulate hydrophobic drugs like N-desmethyltamoxifen, enabling a synergistic combination of phototoxicity and adjuvant chemotherapy.
