Overview
The **prototype oral ETEC vaccine** is an early-generation, oral, inactivated whole-cell vaccine candidate developed by the Gothenburg University Vaccine Research Institute (GUVAX) at the University of Gothenburg, Sweden. Designed to prevent enterotoxigenic *Escherichia coli* (ETEC) diarrhea—a leading cause of diarrheal illness in children in low- and middle-income countries and in international travelers—the vaccine consists of inactivated recombinant *E. coli* strains overexpressing key ETEC colonization factors (CFA/I, CS3, CS5, and CS6) along with the hybrid heat-labile toxoid LCTBA. This formulation targets the bacterial colonization factors and enterotoxins responsible for ETEC-induced diarrhea. The prototype served as a precursor to the more advanced ETVAX vaccine and has been evaluated in a completed Phase I clinical trial in healthy adult volunteers in Sweden to assess safety and immunogenicity.
