Proviza is an investigational small molecule benzodiazepine prodrug being developed by Sollievo Pharmaceuticals for the rapid sedation of acutely agitated and violent patients, including those experiencing acute alcohol withdrawal. It consists of avizafone, a highly water-soluble prodrug that undergoes rapid enzymatic conversion in the bloodstream to diazepam. Formulated for intramuscular injection, Proviza is designed to provide a faster onset of action and more predictable pharmacokinetics than current standards of care like lorazepam, with a median time to peak concentration four times faster and significantly reduced patient-to-patient variability. It typically has an intermediate duration of action lasting between 2 and 4 hours.
