Overview

Proximicin A is a novel aminofuran natural product and research compound isolated from marine strains of the actinomycete *Verrucosispora*. It is a small molecule heterocyclic peptide characterized by the presence of 4-amino-furan-2-carboxylic acid, making it structurally analogous to netropsin and distamycin. Proximicin A exhibits potent cytostatic and antitumor activity against various human cancer cell lines, including gastric adenocarcinoma, hepatocellular carcinoma, breast carcinoma, and glioblastoma. Its mechanism of action involves selective binding to AT-rich DNA sequences in the minor groove, which induces cell cycle arrest at the G1/S phase transition and triggers apoptosis. Additionally, it has been shown to interact with receptors like HER2. Currently, it remains an investigational compound studied primarily in academic settings for its potential as a broad-spectrum anticancer agent.
