PRS-050-PEG40 (also known as Angiocal) is a pegylated Anticalin protein therapeutic engineered to target and antagonize vascular endothelial growth factor A (VEGF-A). It is based on human tear lipocalin and was developed using combinatorial phage display technology. The drug binds specifically to VEGF-A, thereby inhibiting its interaction with VEGF receptors—primarily VEGF receptor 2 (KDR/VEGFR2)—and blocking downstream signaling pathways involved in angiogenesis. This mechanism confers potential antineoplastic activity by suppressing tumor blood vessel formation. The PEGylation with a 40 kDa polyethylene glycol moiety extends the molecule’s half-life and reduces renal clearance. Preclinical studies demonstrated efficacy in xenograft models, and phase I clinical trials evaluated safety and pharmacokinetics in patients with advanced solid tumors[1][2][5].
