PRS-055 is a mono-specific Anticalin protein designed to target and activate CD137 (4-1BB), a co-stimulatory receptor expressed on the surface of activated T cells and natural killer (NK) cells. Developed by Pieris Pharmaceuticals, PRS-055 utilizes the proprietary Anticalin technology platform, which involves engineering human lipocalins to bind specific targets with high affinity and selectivity. By binding to CD137, PRS-055 aims to enhance the anti-tumor immune response by promoting T cell proliferation and survival. While initially developed as a standalone agonist, the program's insights and the CD137-binding Anticalin component were subsequently integrated into the development of Pieris's bispecific Anticalin-antibody fusion proteins (DuoCalins), such as PRS-343 (CD137 x HER2).
