Overview

PRS-342 is a bispecific antibody fusion protein designed for immuno-oncology applications. It consists of an anti-GPC3 antagonistic antibody linked to a 4-1BB (CD137) agonistic Anticalin protein. The drug binds with high affinity to cells expressing both human GPC3 and 4-1BB, targeting GPC3-positive tumor cells and activating 4-1BB on tumor-infiltrating immune cells. This dual mechanism aims to provide potent costimulatory signals that enhance T-cell mediated cytotoxicity specifically within the tumor microenvironment, leading to increased T-cell infiltration and tumor cell killing. PRS-342 was originally developed by Pieris Pharmaceuticals using their proprietary Anticalin technology platform and has been licensed exclusively worldwide to Boston Pharmaceuticals for further development[1][2][3][5][7].
