PRS-400 is an inhaled anti-Jagged-1 Anticalin protein therapeutic being developed by Pieris Pharmaceuticals for the treatment of muco-obstructive lung diseases, including asthma, chronic obstructive pulmonary disease (COPD), and cystic fibrosis. The drug acts by inhibiting Jagged-1, a ligand in the Notch signaling pathway that regulates cell fate specification in the lung epithelium. By blocking Jagged-1-induced Notch2 activation, PRS-400 prevents the differentiation of progenitor cells into mucus-secreting goblet cells and instead promotes ciliated cell formation. This mechanism reduces mucus hypersecretion and addresses airway obstruction at its root cause. The therapy is designed for local delivery to the lungs via inhalation to minimize systemic exposure and associated side effects[1][2][3][6][9].
