PRS-639058 is a high-affinity Anticalin protein that specifically targets Glypican-3 (GPC3), a cell-surface oncofetal antigen frequently overexpressed in hepatocellular carcinoma (HCC) and other solid tumors. Developed by Pieris Pharmaceuticals using their proprietary Anticalin technology, PRS-639058 is an engineered version of a human lipocalin protein. It is primarily utilized as the GPC3-targeting component of the bispecific Anticalin PRS-342 (also known as GPC3-4-1BB), which is designed to provide localized co-stimulation of T cells via 4-1BB (CD137) only in the presence of GPC3-expressing tumor cells. By binding to GPC3, PRS-639058 facilitates the localized activation of the immune system within the tumor microenvironment, potentially reducing systemic toxicities associated with 4-1BB agonism while enhancing anti-tumor efficacy.
