Overview

PRS Regimen V is a novel, ultrashort combination drug regimen designed for the treatment of multidrug-resistant tuberculosis (MDR-TB) and extensively drug-resistant tuberculosis (XDR-TB). It consists of four drugs: bedaquiline, delamanid, clofazimine, and pyrazinamide. Developed using the parabolic response surface (PRS) platform to optimize efficacy and minimize treatment duration, this regimen aims to significantly shorten therapy time compared to standard MDR-TB regimens. Preclinical studies in mice demonstrated that PRS Regimen V rapidly sterilizes lung infections and achieves relapse-free cure rates equivalent to or better than other benchmark regimens. The combination notably excludes fluoroquinolones and aminoglycosides, reducing toxicity risks associated with these classes[3][6]. Clinical trials are ongoing to evaluate its safety and efficacy in humans[1][3][10].
