Drug intelligence / Profile preview
PRT-101 is a first-in-class small-molecule SOX9 transcriptional condensate modulator being developed for the treatment of osteoarthritis. Discovered using Proteina's proprietary Single-molecule Protein Interaction Detection (SPID) platform, PRT-101 directly binds to an intrinsically disordered region of the SRY-box transcription factor 9 (SOX9), which is a master regulator of cartilage-specific gene expression. This binding promotes biomolecular condensate formation (molecular clustering), enhancing SOX9 transcriptional activity and selectively upregulating cartilage-forming genes such as Type II collagen (COL2A1) and aggrecan (ACAN). In preclinical models of severe osteoarthritis, PRT-101 has demonstrated significant efficacy in cartilage regeneration, structural maintenance, pain relief, and reduction of joint inflammation.
