PRT-2761 is a small molecule inhibitor of spleen tyrosine kinase (Syk), developed initially by Portola Pharmaceuticals and later in collaboration with Ora. It was investigated as a topical ophthalmic solution for the treatment of acute and chronic allergic conjunctivitis. The drug acts by inhibiting Syk, a key enzyme involved in mast cell activation and allergic inflammatory responses, thereby reducing symptoms such as ocular itch and redness. Clinical trials reached Phase 2, but development for allergic conjunctivitis was discontinued before approval. There were also considerations to pursue indications in dry eye disease and other ocular inflammatory conditions[1][2][3][5][7].
