PRT1419 is a potent, selective, and orally bioavailable small molecule inhibitor of the anti-apoptotic protein myeloid cell leukemia 1 (MCL1), a member of the BCL2 family. MCL1 plays a critical role in maintaining cellular homeostasis and promoting cancer cell survival, and is frequently overexpressed in both solid tumors and hematologic malignancies. By binding to and inhibiting MCL1, PRT1419 induces apoptosis in cancer cells. The drug was developed by Prelude Therapeutics for use as an antineoplastic agent in various cancers, including relapsed/refractory hematologic malignancies (such as acute myeloid leukemia, chronic myelomonocytic leukemia, high-risk myelodysplastic syndrome) as well as advanced solid tumors (including breast cancer, melanoma, esophageal carcinoma). Clinical development included Phase 1/2 trials; however, development for hematological malignancies and solid tumors has been discontinued[1][4][6][7].
