Overview

PRT2527 + zanubrutinib is an investigational **combination therapy** uniting PRT2527—a potent and highly selective cyclin-dependent kinase 9 (CDK9) inhibitor—with zanubrutinib, a Bruton tyrosine kinase (BTK) inhibitor. The combination is being evaluated for the treatment of relapsed or refractory hematologic malignancies, especially lymphoid cancers. PRT2527 functions by inhibiting CDK9, leading to transcriptional repression of oncogenic drivers such as MYC and MCL1, while zanubrutinib targets and blocks BTK signaling critical for B-cell malignancy survival. The regimen has demonstrated preliminary anti-tumor activity and tolerable safety, with neutropenia as the most common treatment-related adverse event. Both agents are administered as follows: PRT2527 by intravenous infusion once weekly, zanubrutinib by oral capsule either once or twice daily[1][2][3][4][5][6].
