Overview

PRT382 is a small molecule inhibitor of protein arginine methyltransferase 5 (PRMT5) developed by Prelude Therapeutics. PRMT5 is an enzyme that catalyzes the symmetric arginine dimethylation (me2s) of both histone and non-histone proteins, acting as an oncogenic driver in various malignancies, including mantle cell lymphoma (MCL). PRT382 works by modulating the cellular chromatin landscape and altering transcriptional profiles, which can sensitize cancer cells to apoptosis. Specifically, research has demonstrated that PRT382 creates vulnerabilities to BH3 mimetics (such as navitoclax or PRT1419) by shifting the apoptotic priming state of the mitochondria, suggesting potential for synergistic combination therapies in relapsed or refractory MCL.
