**PRTH-101 + pembrolizumab** is an investigational combination regimen for advanced or metastatic solid tumors. **PRTH-101** is a humanized monoclonal IgG1 antibody that targets and inhibits discoidin domain receptor 1 (DDR1), a collagen-binding protein and kinase present on epithelial cells. DDR1 is implicated in forming a physical collagen barrier that excludes immune cells from the tumor microenvironment. By allosterically disabling DDR1, PRTH-101 disrupts collagen alignment, enabling immune cell infiltration into tumors. This leads to increased lymphocyte infiltration and tumor growth inhibition. **Pembrolizumab** (KEYTRUDA) is a humanized monoclonal IgG4 antibody checkpoint inhibitor that binds to and inhibits programmed cell death protein 1 (PD-1) on T cells, restoring antitumor immunity. Combination therapy aims to synergistically increase T-cell entry into tumors and enhance immune-mediated tumor cell killing. Clinical trials are ongoing with this regimen in multiple tumor types, including thymic, colorectal, pancreatic, ovarian, glioma, and non-small cell lung cancer[1][3][5].
