Overview

Pru p 3 is a 9 kDa nonspecific lipid transfer protein (nsLTP) that serves as the primary allergen in peaches (*Prunus persica*). In Mediterranean populations, it is a major sensitizer responsible for severe food-allergic reactions, including anaphylaxis. As a therapeutic agent, Pru p 3 is utilized in allergen-specific immunotherapy (AIT), particularly via sublingual administration (SLIT), to induce clinical desensitization and long-term immune tolerance. The mechanism of action involves modulating the immune system toward a tolerogenic profile, characterized by epigenetic modifications such as DNA methylation changes in dendritic cells (DCs). These alterations regulate the expression of genes involved in T cell activation, antigen presentation, and inflammatory signaling, including HLA-DQA1, HLA-DQB1, FOXP1, and MTOR.
