PRV-3279 is a humanized bispecific monoclonal antibody (diabody, also described as a DART molecule) that targets the B cell surface proteins CD32B and CD79B. By simultaneously engaging these two receptors, PRV-3279 inhibits B cell function and suppresses autoantibody production without causing B cell depletion. This mechanism allows for the regulation of pathogenic B cells while sparing non-pathogenic ones, potentially offering a safer alternative to existing B-cell-targeted therapies. The drug is being developed primarily for systemic lupus erythematosus (SLE), but it may also have applications in other autoimmune diseases where aberrant B cell activity plays a role, as well as in preventing immunogenicity of gene therapies and biotherapeutics. The original developer was MacroGenics; development has been led by Provention Bio (now part of Sanofi), with Hangzhou Zhongmei Huadong Pharmaceutical collaborating for Greater China[1][2][3][4][5][6][7].
