**PRX-034** is a small-molecule **TLR7 agonist** developed by Primmune Therapeutics for oncology and infectious diseases. It preferentially activates plasmacytoid dendritic cells (pDCs) to induce a robust interferon (IFN) response, including IFNα, with minimal proinflammatory cytokines like TNFα and IL-6, enabling systemic immune activation suitable for combination with immune checkpoint inhibitors (ICPIs). PRX-034 binds the guanosine site of TLR7, shows higher potency in human PBMCs (EC50 15 µM for pDC activation) than monkey cells, and is delivered systemically via oral prodrug PRTX007 after hepatic conversion.[1][3][9][11][14]
