PRX005 is a humanized monoclonal tri-epitopic antibody that specifically binds with high affinity to the R1, R2, and R3 repeats within the microtubule binding region (MTBR) of tau protein. It targets both 3R and 4R tau isoforms and is designed to reduce pathogenic tau uptake into neurons. PRX005 is being developed for the treatment of Alzheimer's disease, where tau proteins can accumulate and form neurofibrillary tangles in the brain. In preclinical studies, PRX005 has demonstrated superior ability to bind, intercept and block cellular internalization of pathogenic tau, and mitigate downstream neurotoxicity compared to other anti-tau antibodies.
