Overview

PRX012 is an investigational next-generation monoclonal antibody designed to treat Alzheimer’s disease by targeting aggregated forms of amyloid beta (Aβ), including protofibrils and plaques. It is engineered for subcutaneous administration, offering a patient-friendly dosing schedule with infrequent, low-volume injections. Preclinical studies have demonstrated that PRX012 binds to both pyroglutamate-modified and unmodified Aβ plaques with high avidity, enabling effective plaque occupancy and clearance at lower doses optimal for subcutaneous delivery. The antibody neutralizes toxic Aβ aggregates and promotes their removal from the brain, potentially slowing neurodegeneration associated with Alzheimer’s disease. Developed by Prothena, PRX012 has received FDA Fast Track designation and is currently in Phase 1 clinical trials[1][2][3][5][7].
