Overview

PRX012-TfR3 is a preclinical-stage monoclonal antibody being developed by Prothena for the treatment of Alzheimer's disease. It is designed to target the N-terminus of amyloid-beta (Aβ) plaques in the brain. To overcome the limitations of the blood-brain barrier (BBB), the molecule incorporates a transferrin receptor (TfR)-binding domain, which facilitates receptor-mediated transcytosis into the central nervous system. This 'brain shuttle' technology is intended to enhance the brain-to-plasma ratio of the antibody, potentially leading to greater plaque reduction and improved clinical outcomes compared to conventional anti-Aβ antibodies.
