PRX123 is a wholly-owned, investigational dual-target peptide vaccine developed by Prothena for the treatment and prevention of Alzheimer’s disease. It is designed to simultaneously target key epitopes within the N-terminus of amyloid beta (Aβ) and the microtubule binding region (MTBR) of tau protein. The vaccine aims to promote clearance of Aβ plaques and block transmission of pathogenic tau, addressing two central pathological features in Alzheimer’s disease. Preclinical studies have shown that PRX123 generates robust immunogenic responses, producing antibodies capable of binding both Aβ plaques and tau tangles in brain tissue, leading to significant reduction in cortical Aβ plaques in animal models. The U.S. FDA has cleared its IND application and granted Fast Track designation for Alzheimer's disease; Phase 1 clinical trials are scheduled for 2024[1][2][4][6][9][10].
