PS-1145 is a **small molecule inhibitor** that specifically targets the **IκB kinase (IKK) complex**, a key upstream activator in the canonical **nuclear factor kappa-B (NF-κB) pathway**[1][3][4]. By inhibiting IKK, PS-1145 blocks the phosphorylation and degradation of IκB, thereby preventing the nuclear translocation and transcriptional activity of NF-κB. This results in reduced transcription of NF-κB target genes, many of which are implicated in inflammation, cell survival, and tumorigenesis[1][3][2][4]. PS-1145 was originally derived from a β-carboline natural product and has an IC50 in the nanomolar range for IKK inhibition[1][4]. It has demonstrated selective anti-proliferative and pro-apoptotic effects in multiple cancer cell lines, including nasopharyngeal carcinoma, diffuse large B cell lymphoma, pancreatic cancer, and bladder cancer[1][3][5][2]. It can sensitize some tumors to standard therapies that activate death signaling, such as TRAIL[5]. Clinical development of PS-1145 remains at the preclinical or early investigational stage, with no known marketing approval[4][6].
