Overview

PS-6130 is a cytidine nucleoside analog that serves as a potent and selective inhibitor of the Hepatitis C Virus (HCV) NS5B RNA-dependent RNA polymerase. Developed through a collaboration between Pharmasset and Roche, the compound is phosphorylated within host cells to its active 5'-triphosphate metabolite. This active form competes with cytidine triphosphate for incorporation into the growing viral RNA strand by the NS5B polymerase, resulting in premature chain termination and the inhibition of viral replication. Although PS-6130 demonstrated significant antiviral activity in vitro, its clinical development was largely superseded by its prodrug derivatives, such as mericitabine (RG7128) and RG1626, which were designed to enhance oral absorption and systemic exposure.
