Drug intelligence / Profile preview
PS003biv is a bivalent, llama-derived single-domain antibody (sdAb or nanobody) that acts as an agonist of protein S (PS). Developed by researchers at INSERM and collaborating academic institutions, PS003biv specifically targets the sex hormone-binding globulin (SHBG)-like domain of protein S. This binding enhances the activated protein C (APC)-cofactor activity of protein S, stimulating the APC-mediated proteolytic inactivation of activated factor V (FVa) without affecting physiological hemostasis. Beyond its anticoagulant properties, PS003biv also promotes the non-anticoagulant, prophagocytic (efferocytotic) activity of protein S, which facilitates the clearance of pathological phosphatidylserine-expressing red blood cells (eryghosts) by macrophages. PS003biv is being investigated in preclinical models as a potential therapeutic agent for sickle cell disease (SCD) to alleviate vaso-occlusive crises, vascular congestion, hemolysis, and thromboinflammation, as well as a safe antithrombotic agent.
