PSA:154–163(155L) peptide is a synthetic, modified agonist peptide derived from the prostate-specific antigen (PSA), specifically corresponding to amino acids 154 to 163 with a leucine substitution at position 155. This modification increases its binding affinity to HLA-A2 molecules. The peptide is designed as a cancer vaccine candidate for prostate cancer, aiming to elicit cytotoxic CD8+ T-cell responses against tumor cells expressing endogenous PSA. It has been evaluated in phase II clinical trials in HLA-A2 positive patients with recurrent or rising serum PSA after radical prostatectomy for prostate cancer. While the vaccine induced CD8+ T-cell responses against both the modified and native peptides in vitro, it failed to induce effective immune recognition of tumor targets expressing endogenous PSA and did not significantly affect serum PSA levels[1][2][4][6].
