PSB-234 is an antibody-targeted immuno-cytokine consisting of an IL-2 mutein conjugated to a targeting antibody. The IL-2 mutein component is engineered with a significantly reduced affinity (>100-fold) for the IL-2 receptor in its unconjugated form, ensuring that signaling is only restored to wild-type levels upon cell-specific anchoring by the antibody. This mechanism allows for the selective expansion and activation of suppressive regulatory T cells (Tregs) while minimizing the expansion of effector immune cells such as CD8+ T cells or NK cells. Developed by Sana Biotechnology, PSB-234 is being investigated for the treatment of Type 1 diabetes (T1D) to restore immune homeostasis and delay the onset of hyperglycemia by protecting pancreatic islets from autoreactive T cell destruction.
