Overview

PSCA gamma-delta CAR T-cell therapy is an investigational autologous cell therapy developed by Moffitt Cancer Center for the treatment of metastatic castration-resistant prostate cancer (mCRPC), particularly for patients with bone metastases. This therapy is distinguished by its use of gamma-delta (γδ) T cells, a specific T-cell subtype that typically constitutes less than 5% of peripheral blood T cells. These cells are harvested from the patient and genetically engineered to express a chimeric antigen receptor (CAR) targeting the prostate stem cell antigen (PSCA), a glycoprotein highly expressed in advanced prostate cancer. The therapy aims to leverage the unique ability of γδ T cells to infiltrate solid tumors and their CAR-independent anti-tumor activities, which may be further enhanced by bone-modifying agents like zoledronic acid. It is currently being evaluated in a Phase 1 clinical trial (NCT06193486) as an intravenous infusion following lymphodepleting chemotherapy.
