PSCA-RevTM is a recombinant targeting module (TM) designed for use with the RevCAR (Reversible Chimeric Antigen Receptor) T-cell platform, a modular immunotherapy system developed to enhance the safety and control of CAR-T cell treatments. The PSCA-RevTM molecule is a bispecific adapter that simultaneously binds to the Prostate Stem Cell Antigen (PSCA) expressed on the surface of cancer cells and to a universal chimeric receptor expressed on engineered RevCAR-T cells. By bridging these two entities, the module facilitates the formation of an immunological synapse, leading to T-cell activation and the targeted destruction of PSCA-positive tumor cells, such as those found in prostate and pancreatic cancers. A key feature of this system is its reversibility; because the RevCAR-T cells are inactive without the targeting module and the module itself has a short biological half-life, the therapeutic effect can be rapidly titrated or terminated by adjusting the module's infusion, thereby providing a safety switch against potential toxicities like cytokine release syndrome (CRS).
