pSD11-Livin1 is an experimental gene therapy construct designed to silence the expression of the Livin gene (also known as BIRC7 or ML-IAP) through RNA interference. It consists of a pSD11 plasmid vector that expresses a specific short hairpin RNA (shRNA) sequence targeting the mRNA of Livin, a member of the Inhibitor of Apoptosis Protein (IAP) family. Livin is frequently overexpressed in various malignancies, such as bladder cancer, lung cancer, and melanoma, where it functions to inhibit programmed cell death by neutralizing caspases. By delivering shRNA to tumor cells, pSD11-Livin1 aims to downregulate Livin protein levels, thereby restoring apoptotic signaling, inhibiting tumor cell proliferation, and potentially increasing the efficacy of concurrent chemotherapeutic agents. Research on this construct has been primarily conducted in preclinical models by academic institutions.
