pSD11-Livin2 is an experimental gene therapy construct designed to suppress the expression of Livin (BIRC7), a member of the Inhibitor of Apoptosis Protein (IAP) family. The construct utilizes the pSD11 eukaryotic expression plasmid vector to deliver short hairpin RNA (shRNA) specifically targeting the Livin isoform 2 (also known as Livin β). Livin is frequently overexpressed in various malignancies, including bladder cancer, melanoma, and colorectal cancer, where it functions to inhibit apoptosis by binding to and neutralizing caspases-3, -7, and -9. By utilizing RNA interference (RNAi) to silence Livin expression, pSD11-Livin2 aims to sensitize cancer cells to programmed cell death, inhibit tumor proliferation, and potentially overcome chemoresistance. In preclinical research, this construct is often delivered using attenuated *Salmonella typhimurium* (such as strain VNP20009) as a live bacterial vector, which selectively colonizes the hypoxic and necrotic regions of solid tumors.
