pSG.SS.Flk-1ECD.C3d3.YL is an experimental DNA vaccine designed for anti-angiogenic cancer immunotherapy. The vaccine consists of a recombinant plasmid encoding the extracellular domain (ECD) of Flk-1 (the murine homolog of Vascular Endothelial Growth Factor Receptor 2, or VEGFR2) fused to three copies of the complement component C3d (C3d3). C3d serves as a molecular adjuvant that bridges innate and adaptive immunity by binding to complement receptor 2 (CR2/CD21) on B cells, thereby significantly enhancing the antibody response against the Flk-1 antigen. By inducing high titers of Flk-1-specific antibodies, the vaccine aims to inhibit VEGF-mediated signaling in vascular endothelial cells, leading to the suppression of tumor angiogenesis and the inhibition of solid tumor growth, as demonstrated in preclinical models of bladder transitional cell carcinoma.
