Overview

pshRNA-survivin is a gene silencing therapeutic utilizing **plasmid-mediated short hairpin RNA (shRNA) designed to target and downregulate the mRNA** of the human gene survivin (also known as BIRC5). Survivin is a member of the inhibitor of apoptosis proteins (IAP) family and is highly expressed in many cancers but not in most normal adult tissues. Its inhibition leads to increased tumor cell apoptosis (via activation of caspase-9 and disruption of cell cycle progression) and reduced cell proliferation. The mechanism of action involves **RNA interference**: the delivered shRNA forms double-stranded RNA that is processed into siRNA by cellular machinery, resulting in specific survivin mRNA cleavage and knockdown. This results in the induction of apoptosis and cell-cycle arrest in tumor cells. pshRNA-survivin is under preclinical and early clinical investigation as an **anticancer genetic therapy**, with particular research focus in non-small cell lung cancer and other solid tumors[6].
