PSI-352938 is a nucleoside derivative and cyclic phosphate prodrug of β-D-2′-deoxy-2′-α-fluoro-2′-β-C-methylguanosine 5′-monophosphate. It acts as a nucleotide analog inhibitor targeting the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase. The drug demonstrates potent antiviral activity against multiple HCV genotypes in vitro and retains activity against replicons with the S282T mutation, which confers resistance to some other nucleoside/nucleotide analogs. PSI-352938 must be metabolized intracellularly to its active triphosphate form (PSI-352666) to inhibit viral replication[7][10][4]. The compound was developed for the treatment of chronic hepatitis C infection and has been evaluated in clinical trials as monotherapy and in combination regimens[6][3].
