Overview

PSIL-002 is a proprietary, non-hallucinogenic analogue of N,N-dimethyltryptamine (DMT) being developed by Psilera as a next-generation psychedelic-inspired neuromodulator for mood and substance use disorders.[1][3][5][7][9] Preclinical in vivo studies in mice show that PSIL-002 is safe and well tolerated across a wide dose range (0.5–100 mg/kg) and does not induce the head-twitch response, a standard behavioral proxy for 5‑HT2A‑mediated hallucinogenic activity, while maintaining antidepressant-like efficacy in the forced swim test comparable to psilacetin (4‑AcO‑DMT).[1][5][7] Additional repeat‑dose studies indicate improvements in learning and memory over psilacetin, suggesting potential utility in neurodegenerative conditions such as dementia and Alzheimer’s disease.[7][9] PSIL-002 arises from Psilera’s patent-pending NCE library, which leverages computational screening (BRAIN platform) to optimize classical tryptamines for non-hallucinogenic, outpatient-use profiles by modulating activity at serotonergic and related targets.[1][5][7]
