PSIL-006

PSIL-006 is a novel, non-hallucinogenic psilocybin derivative developed by Psilera for the treatment of neurodegenerative and neuropsychiatric disorders. It is designed as a next-generation "neuroplastogen" with a dual agonist mechanism at serotonin 2A (5-HT2A) and serotonin 6 (5-HT6) receptors. Unlike traditional psychedelics, PSIL-006 selectively reduces hallucinogenic effects while retaining therapeutic activity, making it suitable for take-home use and potentially broadening patient access. Preclinical studies have demonstrated that PSIL-006 improves learning, memory, and sleep in humanized Tau mouse models relevant to frontotemporal dementia (FTD), with no limiting side effects observed. The lead indication is behavioral variant frontotemporal dementia (bvFTD), an early-onset form of dementia with significant unmet medical need[1][4][5][7]. Additional preclinical development has explored its potential in alcohol use disorder, anxiety disorders, cognition disorders, and depressive disorder[3].
